This kit is used to measure the 5’-Nucleotidase (5’-NT) activity in serum and plasma samples.

Detection principle 

The activity of 5’-NT in the sample can be calculated according to measure the generation rate of quinone compounds at 546 nm.

Sample preparation

Collect fasting blood and separate serum or plasma by routine method. The samples can be store at 4℃ for 4 days.

Operation steps

Mix thoroughly, incubate at 37℃ for 3 min. Measure the absorbance at 546 nm at 0 second (A1) and 120 second (A2), respectively. Calculate the △A/min.

Reference range

Serum and plasma: 2-11.4U/L.

This is for reference only. It is recommended to establish its own range of reference values.

Limitations of test method

Interfering substance: hemoglobin ≤ 10 g/L, conjugated bilirubin ≤ 3 mg/dL, unconjugated bilirubin ≤ 100 mg/dL, VC ≤ 11.25 mg/dL, TG ≤ 816 mg/dL have no interference for detection.

Technical parameter

1. The absorbance of blank tube is ≤ 0.3 (Optical path=1.0 cm).

2. Linear range: 1-300U/L, r2 ≥ 0.995.

3. Accuracy: Relative deviation ≤ 10%.

4. Repeatability: intra-assay CV ≤ 6.0%, inter-assay CV ≤ 10.0%.

5. Sensitivity: when test the sample (the concentration of sample is 60U/L), △A is ≥ 0.01.

6. Storage: The valid of kit is 12 months when stored at 4℃ in the dark. It is stable for 1 month when stored at 4℃ in the dark after opening.

Notes

1. This product is for scientific research use only.

2. Do not use components from different batches of kit.

3. Avoid to contact the reagent with skin and clothing. Wash immediately with plenty of water if contact it carelessly
